The FLAG-tag (DYKDDDDK) is a short polypeptide added to proteins via recombinant DNA technology. It is widely used for protein purification by affinity chromatography and for studying proteins in living cells. The tag helps isolate recombinant proteins from host proteins and enables purification of multi-subunit complexes without disrupting them. It is also used to obtain proteins suitable for x-ray crystallography. The FLAG-tag can be detected with specific antibodies, making it useful in assays like immunofluorescence, SDS-PAGE, and Western blotting, especially when no antibody exists for the target protein.
